Pikovskaya's Agar

• This media is generally available as a premade powder.

Step 1

• Fill a 2L flask or bottle with 1000 ml DI-H~2~O
• Weigh out ingredients and add to water in container
  • Yeast extract - 0.5 g
  • Dextrose - 10.0 g
  • Tri-calcium phosphate - 5.0 g
  • Ammonium sulfate - 0.5 g
  • Potassium chloride - 0.2 g
  • Magnesium sulfate - 0.1 g
  • Manganese sulfate - 0.0001 g
  • Ferrous sulfate - 0.0001 g

Step 2

• Loosely screw on cap to bottle
  • or cover opening of flask with aluminum foil
• Mark with autoclave tape
• Autoclave media at 121^o^C and 15 psi for 25 mins
• After autoclaving allow media to cool to ~ 65^o^C
• Dispense 15 ml media into 100 mm petri plates
• Cover plates and allow to dry overnight
• Package plates into bags and tape closed
• Place on shelf to store for later
